Default Is it the salt or the water?

What would happen if you slipped a seastone into Luffy's food? I doubt he'd notice but what would the effects be. Also, are DF users affected by a fresh water lake? Small amounts of just salt water doesn't seem to affect DF users since their own sweat would defeat them and that would be pretty lame.

Any large body of standing water, either salt water or fresh water will rob them of their powers, but they have to be submerged to not be able to use their powers they can still be around it. And if they are half submerged (they must be in a shallow area) then they can use their powers using body parts above the water, they can't use their powers under water, but anybody else can manipulate their bodies if it has been changed as shown during the Arlong fight when they stretched Luffy's head. Any running water won't effect them (waves, rain). And seastones only work with direct and constant contact, so Luffy would notice that he wouldn't be able to use his powers and depending on the density and composition he might not be able to move around until it passed through his system and exited his body.

too bad seastone is so rare...
i'd powder it and put it in an exploding canonball
covering the entire vessel and crew would be a good way to start the battle...
what would happen to Brooke in contact with seastone, would he be dead for a bit... chitchat with st. peter and popback once the stone was removed?
would chopper just be a normal blue nosed reindeer with no human intelligence?

They can use thier powers even in water, it is just when they are in water they lose a lot of their power and any kind of task feels much heavier on them, thats why they are called "hammers". That or becuase most would sink to the bottom of the sea and drown.
